Uses of Stalevo Tablet

• Manage Parkinson’s disease

Composition:

Each tablet contains a 1:4 ratio of carbidopa to levodopa and 200 mg of entacapone. Available in various strengths:

• Stalevo 50: 12.5 mg carbidopa, 50 mg levodopa, 200 mg entacapone
• Stalevo 75: 18.75 mg carbidopa, 75 mg levodopa, 200 mg entacapone
• Stalevo 100: 25 mg carbidopa, 100 mg levodopa, 200 mg entacapone
• Stalevo 125: 31.25 mg carbidopa, 125 mg levodopa, 200 mg entacapone
• Stalevo 150: 37.5 mg carbidopa, 150 mg levodopa, 200 mg entacapone
• Stalevo 200: 50 mg carbidopa, 200 mg levodopa, 200 mg entacapone

Side Effects of Stalevo

• Common: Dyskinesia, nausea, hyperkinesia, urine discoloration, diarrhea, stomach pain.
• Severe: Severe diarrhea, colitis, hallucinations, orthostatic hypotension, rhabdomyolysis, neuroleptic malignant syndrome, skin cancer.
• Dose Adjustments: Based on clinical response; may require reducing levodopa dose if dyskinesia occurs.
• Overdose Management: Includes agitation, confusion, bradycardia, and supportive measures such as gastric lavage and charcoal.

• You have been prescribed Stalevo to treat your symptoms of Parkinson's disease.
• Avoid a high-protein diet (eg. meat, poultry, and dairy products) while you are taking this medicine as that can reduce the amount of medication absorbed by your body.
• It may cause dizziness or sleepiness. Don't drive or do anything requiring concentration until you know how it affects you.
• To lower the chance of feeling dizzy or passing out, rise slowly if you have been sitting or lying down.
• Do not stop taking the medication suddenly without talking to your doctor. It may result in a serious syndrome that causes fever, stiff muscles, unusual body movements, sweating, and confusion.
• Talk to your doctor in case you develop hallucinations or notice other changes in your behavior.

Indications of Stalevo

• Treatment of adult patients with Parkinson's disease experiencing end-of-dose motor fluctuations not stabilized on levodopa/DDC inhibitor treatment.

Pharmacology

• Levodopa: Converted to dopamine in the brain to alleviate Parkinson's symptoms.
• Carbidopa: Inhibits peripheral metabolism of levodopa, increasing its availability to the brain.
• Entacapone: Inhibits catechol-O-methyltransferase (COMT), enhancing and prolonging the effect of levodopa.

Dosage & Administration of Stalevo

• Adults: Dosage should be individually titrated. One tablet per dose administration. Maximum entacapone dose: 2,000 mg/day.
• Take orally, with or without food, as whole tablets.

Interaction

• Antihypertensives: May require dose adjustment due to the risk of hypotension.
• Dopamine antagonists, phenytoin, and papaverine: May reduce levodopa efficacy.
• High Protein Diet: May impair levodopa absorption.

Contraindications

Severe hepatic impairment, narrow-angle glaucoma, pheochromocytoma, MAO inhibitors, history of NMS, or hypersensitivity to components.

Pregnancy & Lactation

Pregnancy category C. The levodopa/carbidopa/entacapone combination should not be used during pregnancy unless the potential benefit justifies the potential risk to the fetus. The safety of this combination in nursing infants is unclear. During the use of this combination therapy, women should not breastfeed.
