✔️ How does Clopixol Depot Injection work?

Zuclopenthixol is a typical antipsychotic medicine that works by restoring the balance of certain chemical substances (neurotransmitters) in the brain. It exerts its actions by antagonizing the effects of dopamine by blocking the D1 and D2 dopamine receptors which improves the symptoms of schizophrenia, making it easier to function in everyday life.

• Drowsiness, blurred vision, tachycardia, nausea, dizziness, headache, excitement, postural hypotension, hyperprolactinemia, sexual dysfunction, ECG changes, hyperthermia, extrapyramidal symptoms, urinary frequency or incontinence, tardive dyskinesia.
• Potentially fatal: Neuroleptic malignant syndrome, blood dyscrasias.

✔️ Pharmacology

Zuclopenthixol, a thioxanthene derivative, binds strongly to dopamine D1 and D2 receptors. It also has a high affinity for α1-adrenergic and 5-HT2 receptors, with weaker activity at histamine H1 receptors, and an even lower affinity for muscarinic cholinergic and α2-adrenergic receptors.

✔️ Dosage & Administration of Clopixol Depot

Zuclopenthixol Depot: Requires close supervision after initiation to avoid over-medication or insufficient symptom control. Oral antipsychotics may be needed initially. The maintenance dose is typically 150-300 mg IM every 2-4 weeks, adjusted as needed.

Renal Dose:

• Dose reduction may be needed for renal impairment.

The injection is given into the muscle of the buttock or thigh. Can be taken with or without food.
