Side Effects of Pregadel 50mg Capsule

Common Side Effects:

• Dizziness, somnolence, dry mouth, edema, blurred vision, weight gain, and cognitive issues.

In Pediatric Patients:

• Increased weight and appetite (for partial onset seizures).

Quick Suggestions:

• Maintain a diet rich in fruits, vegetables, whole grains, and omega-3-rich foods. Avoid fried foods, high-fat dairy products, pastries, and processed foods, as they may worsen neuropathic pain.
• Do regular exercise and maintain a healthy weight.
• Avoid the consumption of alcohol as it may increase dizziness or drowsiness.
• Do not take Pregadel 50mg Capsule for a prolonged time; it may lead to mental or physical dependence on Pregadel 50mg Capsule.

Pregadel is prescribed for:

• Neuropathic pain from diabetic peripheral neuropathy (DPN)
• Postherpetic neuralgia (PHN)
• As an adjunctive therapy for partial-onset seizures in patients aged 1 month and older
• Fibromyalgia
• Neuropathic pain associated with spinal cord injury

Pharmacology

Pregabalin, a structural derivative of gamma-aminobutyric acid (GABA), does not bind directly to GABAA, GABAB, or benzodiazepine receptors. It binds with high affinity to the alpha2-delta site (a subunit of voltage-gated calcium channels) in the central nervous system. While the precise mechanism of action is unclear, animal studies suggest that this binding may contribute to its pain-relieving and anti-seizure effects.

Dosage & Administration of Pregadel 50mg Capsule

For Neuropathic Pain Associated with Diabetic Peripheral Neuropathy (DPN) in Adults:

• Immediate Release: Start with 50 mg three times a day (150 mg/day), increasing to 100 mg three times a day (300 mg/day) as needed based on efficacy and tolerability.
• Extended Release (CR): Start with 165 mg once daily, increasing to 330 mg once daily within a week based on patient response and tolerability. The maximum dose is 330 mg once daily.

For Postherpetic Neuralgia (PHN) in Adults:

• Immediate Release: Start with 75 mg twice a day or 50 mg three times a day (150 mg/day), increasing to 150 mg twice a day or 100 mg three times a day (300 mg/day) within a week. The maximum dose is 300 mg twice a day or 200 mg three times a day (600 mg/day).
• Extended Release (CR): Start with 165 mg once daily, increasing to 330 mg once daily within a week. If inadequate pain relief is noted after 2-4 weeks and the patient tolerates the drug, the dose can be increased to 660 mg once daily. The maximum dose is 660 mg once daily.

For Fibromyalgia in Adults:

• Start with 75 mg twice a day (150 mg/day), increasing to 150 mg twice a day (300 mg/day) within a week. If necessary, the dose can be increased to 225 mg twice a day (450 mg/day).

For Neuropathic Pain Associated with Spinal Cord Injury in Adults:

• Start with 75 mg twice a day (150 mg/day), increasing to 150 mg twice a day (300 mg/day) within a week. If necessary, the dose can be increased to 300 mg twice a day (600 mg/day).

Administration

Route of administration: Oral, with or without food.

• Extended Release (CR): Take after an evening meal. Swallow whole, do not split, crush, or chew.

Interaction

• Minimal significant pharmacokinetic interactions.
• No specific interactions with food.

Contraindications

Known hypersensitivity to Pregabalin or its components.

Pregnancy & Lactation

• No adequate studies on pregnant women. Pregnant women should be informed of potential risks.
• Small amounts of Pregabalin were detected in breast milk; breastfeeding is not recommended during treatment due to potential tumorigenicity.

Precautions & Warnings

• Angioedema can occur and may require emergency treatment. Discontinue if hypersensitivity reactions occur.
• Increased risk of suicidal thoughts/behavior.
• May cause dizziness and somnolence; and impair the ability to drive or operate machinery.
• Peripheral edema possible; caution with coadministration of thiazolidinediones.
• Safety and effectiveness for children with neuropathic pain or fibromyalgia have not been established. For adjunctive therapy in partial-onset seizures, safety in patients below 1 month old is unverified. Extended-release tablets' safety in pediatric patients is also not established.
