Side Effects of Vibrenta Penset

Possible side effects include hypoglycemia, allergic reactions, injection site reactions, lipodystrophy, itching, and rashes.

Quick Suggestions:

• Inject subcutaneously (under the skin) in the upper arm, thigh, or abdomen. Rotate injection sites to prevent skin changes at any one site.
• Administer once daily at the same time each day for consistent blood sugar control.
• Regularly monitor blood sugar levels to adjust doses and prevent hypoglycemia.
• Opened vials/cartridges can be stored at room temperature for up to 4 weeks; unopened vials should be refrigerated (2°C–8°C).

Indication of Vibrenta Penset

Vibrenta is prescribed to improve glycemic control in both adults and children with type 1 diabetes mellitus and in adults with type 2 diabetes mellitus.

Pharmacology

Insulin Glargine is a sterile solution used for subcutaneous injection. It is a long-acting (up to 24 hours) human insulin analog produced through recombinant DNA technology. Its primary function is to regulate glucose metabolism. It lowers blood glucose by stimulating peripheral glucose uptake, mainly by skeletal muscle and fat, and by inhibiting hepatic glucose production. Insulin also inhibits lipolysis and proteolysis and enhances protein synthesis.

Dosage & Administration of Vibrenta Penset

Insulin Glargine should be administered once daily at the same time each day. Dosages are personalized based on clinical response, with regular blood glucose monitoring essential. For Type 1 diabetes, it must be combined with short-acting insulin. It is not suitable for intravenous administration as it may cause severe hypoglycemia.

Starting Therapy:

• Type 1 Diabetes: Begin with roughly one-third of the total daily insulin requirement, supplementing with short-acting insulin.
• Type 2 Diabetes (new to insulin): Start with 10 units (or 0.2 units/kg) once daily, adjusting as needed.

Converting from Other Insulin Therapies:

• From once-daily NPH: Begin with the same dose as the NPH being replaced.
• From twice-daily NPH: Start with 80% of the total NPH dose being replaced.

Cartridge: Insert into the pen, attach the needle, and mix the insulin by gently turning the pen upside down 8-10 times. Adjust dosage, remove the needle cap, expel air bubbles, and inject.

Interaction

Various drugs affect glucose metabolism, potentially requiring dose adjustments.

Drugs that may reduce requirements:

• Oral anti-diabetic drugs, ACE inhibitors, disopyramide, fibrates, fluoxetine, monoamine oxidase inhibitors, propoxyphene, pentoxifylline, salicylates, sulfonamide antibiotics.

Drugs that may increase requirements:

• Thiazides, glucocorticoids, thyroid hormones, beta-sympathomimetics, growth hormone, danazol. Beta-blockers, clonidine, lithium, and alcohol may either increase or decrease the effect.

Contraindications

It is not for use in patients hypersensitive to insulin galline or its ingredients.

Pregnancy & Lactation

Classified as Pregnancy Category C. Use during pregnancy only if benefits outweigh risks. The excretion in human milk is unknown, so caution is advised for nursing women, with potential dose and dietary adjustments needed.

Precautions & Warnings

Dose Adjustment and Monitoring: Regular monitoring of blood glucose is necessary. Adjustments should be made under medical supervision.

Administration: Do not dilute or mix with other insulins or solutions. Not for use in insulin pumps or intravenously due to hypoglycemia risk.

Renal or Hepatic Impairment: Dose reductions may be necessary.
