Side Effects of Denixil 0.5mg Tablet

Common side effects include CNS depression, with drowsiness occurring in about 50% of patients, and ataxia in about 30%. Behavioral problems in about 25% of patients. Other side effects: abnormal eye movements, aphonia, coma, tremor, vertigo, confusion, depression, amnesia, hallucinations, hysteria, increased libido, insomnia, psychosis, and palpitations.

How does Denixil 0.5mg Tablet work?

Clonazepam works by stimulating the release of a chemical [gamma-amino-butyric-acid (GABA)] that calms the abnormal electrical activity in the brain.

Quick Suggestions:

• Take Clonazepam with/without food, and take the medicine with a sufficient amount of water.
• Do not break, crush, or chew the tablet/capsules. It should be taken exactly as instructed by your doctor.
• Given that the medicine has habit-forming tendencies, taking the right quantity for the right duration is critical.
• This medicine may cause dizziness, and avoid doing activities (such as driving or operating heavy machines) that require high mental alertness. Never discontinue usage without consulting your doctor.

Indication of Denixil 0.5mg Tablet

• Panic Disorder: Treatment of panic disorder, with or without agoraphobia. Panic disorder is characterized by unexpected panic attacks and concern about having additional attacks or worry about the consequences.
• Seizure Disorders: Alone or as an adjunct in treating Lennox-Gastaut Syndrome (petit mal variant), akinetic, and myoclonic seizures. It may be indicated for patients with absence seizures (petit mal) who have not responded to succinimides.

The effectiveness of Denixil for long-term use (over 9 weeks) has not been systematically studied. Physicians should periodically reevaluate the drug's long-term usefulness for individual patients.

Pharmacology

Clonazepam, a benzodiazepine, has anticonvulsive, sedative, muscle-relaxing, and anxiolytic properties. These effects are mediated through the enhancement of GABAergic neurotransmission at inhibitory synapses, increasing GABA receptor affinity for the neurotransmitter, and enhancing chloride ion flux. Clonazepam also affects serotonin and has been shown to suppress various types of paroxysmal activity, making it beneficial for both generalized and focal epilepsies.

Dosage & Administration of Denixil 0.5mg Tablet

Oral Administration:

• Adults:
  • Seizure disorders: The initial dose should not exceed 1.5 mg/day in three doses. Increase by 0.5 to 1 mg every 3 days until control or side effects preclude further increase. The maximum daily dose is 20 mg.
  • Panic disorder: Initial dose is 0.25 mg in two divided doses, increasing to 1 mg/day after 3 days.
• Pediatric Patients:
  • For infants and children (up to 10 years or 30 kg), the initial dose is 0.01-0.03 mg/kg/day, not exceeding 0.05 mg/kg/day, in two or three divided doses.

Interaction

Denixil does not alter the pharmacokinetics of phenytoin, carbamazepine, or phenobarbital. The effect on other drugs is not investigated.

Contraindications

• Hypersensitivity to benzodiazepines
• Significant liver disease
• Contraindicated in acute narrow-angle glaucoma, but can be used with open-angle glaucoma receiving appropriate therapy.

Pregnancy & Lactation

• Pregnancy: Clonazepam may cause congenital malformations. It should be administered only if the benefits outweigh the risks. High doses during the last trimester of labor can cause irregular fetal heartbeat, hypothermia, hypotonia, mild respiratory depression, and poor feeding in the neonate. Abrupt discontinuation can exacerbate epilepsy.
• Nursing Mothers: Clonazepam passes into maternal milk in small amounts. If necessary, breastfeeding should be discontinued.

Precautions & Warnings

• Coexisting seizure disorders may require additional anticonvulsants.
• Valproic acid with Denixil may cause absence status.
• For pediatric patients, monitor for increased salivation and bronchial secretion.
• Elderly patients are more sensitive to benzodiazepine effects.
• Renal impairment does not require dose adjustment.
• Use caution with hepatic impairment due to altered drug clearance.

Storage Conditions:

Store in a dry place away from light and heat, and keep out of reach of children.
