It works by increasing the action of a chemical messenger (GABA) which suppresses the abnormal and excessive activity of the nerve cells in the brain.
Common side effects include CNS depression, drowsiness (50% of cases), and ataxia (30%). Behavioral problems have been observed in 25% of patients. Other side effects include tremors, confusion, depression, hallucinations, and psychosis.
Overdose may cause drowsiness, ataxia, coma, and respiratory depression. Monitoring and supportive treatment are necessary, and flumazenil may be used as an antidote in severe cases.
Clonazepam enhances GABAergic neurotransmission at inhibitory synapses, thereby increasing GABA’s effect on chloride ion flux at postsynaptic receptors. This leads to the anticonvulsant and sedative effects of the drug. It has also shown effects on serotonin and can suppress various types of paroxysmal EEG activity, making it useful in generalized and focal epilepsies.
Oral:
Leptic does not alter the pharmacokinetics of phenytoin, carbamazepine, or phenobarbital. Its effect on the metabolism of other drugs remains unstudied.
