Doxophylline is a novel bronchodilator structurally related to theophylline but distinguished by the presence of a dioxolane group at position 7.
It acts by selectively inhibiting phosphodiesterase-4 (PDE-4), resulting in bronchial smooth muscle relaxation and improved airflow.
Unlike theophylline, Doxophylline has a lower affinity for adenosine A1 and A2 receptors, which contributes to its better safety and tolerability.
Additionally, it inhibits platelet-activating factor (PAF) and leukotriene synthesis, offering anti-inflammatory benefits.
Adults:
400 mg tablet two or three times daily, or as prescribed by the physician.
Elderly:
200 mg tablet two or three times daily.
Children:
>12 years: 200 mg tablet or 10 ml syrup two or three times daily.
6–12 years: 6–9 mg/kg body weight twice daily (e.g., 3 ml syrup or 60 mg twice daily for a 10 kg child).
For a daily requirement of 400 mg, the Doxophylline SR tablet may be taken once daily.
Nausea, vomiting, epigastric pain
Headache, insomnia, irritability, dizziness
Tachycardia, palpitations, tachypnea
Rarely, hyperglycemia or albuminuria
Known hypersensitivity to Doxophylline or other xanthine derivatives
Reduced hepatic clearance may occur in elderly patients and those with heart failure, COPD, liver disease, or infections — dose adjustment and drug level monitoring are recommended.
Store in a cool, dry place below 30°C, away from light and moisture.
