Palbociclib selectively and reversibly inhibits cyclin-dependent kinases (CDK) 4 and 6. These kinases, along with cyclin D1, play a role in cell proliferation by regulating the cell cycle progression from the G1 to S phase. Inhibiting CDK4/6 slows the proliferation of breast cancer cells.

- Recommended Dose: 125 mg capsule, taken orally once daily for 21 consecutive days, followed by a 7-day break (28-day cycle).
- Administration Instructions:
- Take with food at the same time each day.
- For men undergoing Palbokin with an aromatase inhibitor, treatment with an LHRH agonist may be considered based on clinical guidelines.
- Dose Adjustments for Adverse Reactions:
- Starting Dose: 125 mg/day.
- First Reduction: 100 mg/day.
- Second Reduction: 75 mg/day.
- Discontinue treatment if further dose reduction is required.
