✔️ How does Verigat 2.5 work?

Verigat 2.5 turns on a protein (soluble guanylate cyclase) in the body which relaxes the blood vessels and allows more blood to flow through. As a result, the heart can pump better and doesn't have to work as hard.

✔️ Side Effects of Verigat 2.5

Common side effects include:

• Low blood pressure (hypotension)
• Low red blood cell count (anemia)

✔️ Quick Suggestions:

• Monitor your blood pressure regularly since this medicine may cause low blood pressure.
• Inform your doctor if you are anemic since Vericiguat can cause a decrease in the hemoglobin level.
• If you feel dizzy while taking this medicine, do not drive a vehicle, or operate any machinery.
• Use contraceptives, and do not conceive a child (get pregnant) while taking Vericiguat. Barrier methods of contraception, such as condoms should be used during treatment and for 1 month after you stop treatment with Vericiguat.

✔️ Indication of Verigat 2.5

Verigat is used to reduce the risk of cardiovascular death and hospitalization due to heart failure (HF) in adults with symptomatic chronic HF and an ejection fraction of less than 45%, particularly following a hospitalization or need for outpatient intravenous (IV) diuretics.

✔️ Pharmacology

Vericiguat works by stimulating soluble guanylate cyclase (sGC), an enzyme critical to the nitric oxide (NO) signaling pathway. NO binds to sGC, which promotes the production of cyclic guanosine monophosphate (cGMP), a molecule that helps regulate vascular tone, cardiac contractility, and cardiac remodeling. Heart failure is often associated with decreased NO production and reduced sGC activity, leading to impaired heart and vascular function. Vericiguat increases cGMP levels both independently and synergistically with NO, causing smooth muscle relaxation and vasodilation.

✔️ Dosage & Administration of Verigat 2.5

• Starting Dose: The initial recommended dose of Vericiguat is 2.5 mg once daily with food. The dose can be doubled every 2 weeks to reach the target maintenance dose of 10 mg once daily, based on patient tolerance.
• For patients unable to swallow tablets: The tablet can be crushed and mixed with water, to be taken immediately before administration, as directed by a physician.
• Geriatric Patients: No dose adjustment is needed for elderly patients. No significant differences in safety or efficacy have been observed between patients aged 65 years and older compared to younger adults.
• Hepatic Impairment: No dose adjustment is needed for mild to moderate liver impairment.
• Renal Impairment: No dose adjustment is required in patients with an estimated glomerular filtration rate (eGFR) ≥15 mL/min/1.73 m² who are not on dialysis. However, Vericiguat has not been studied in patients with eGFR <15 mL/min/1.73 m² or those on dialysis.
• Pediatric Use: The safety and efficacy of Vericiguat in children have not been established.

Swallow the tablet as a whole with water. Tablets may be crushed and mixed with water for patients who have difficulty swallowing. Use the medicine as prescribed by a registered physician.

✔️ Interaction

• sGC stimulators: Vericiguat should not be used alongside other soluble guanylate cyclase stimulators.
• PDE-5 Inhibitors: It is recommended to avoid using Vericiguat with PDE-5 inhibitors as it may cause low blood pressure (hypotension).
• Food: Vericiguat should be taken with food for maximum bioavailability, which is 93% when taken with a meal.

✔️ Contraindications

Vericiguat is contraindicated in patients who are also taking other soluble guanylate cyclase stimulators.

✔️ Pregnancy & Lactation

• Pregnancy: Vericiguat is contraindicated during pregnancy due to the risk of harm to the fetus. Females of reproductive potential must use effective contraception during treatment and for one month after stopping treatment.
• Breastfeeding: There is no information regarding the presence of Vericiguat in human breast milk, its effects on the infant, or its impact on milk production. Due to potential risks to the infant, breastfeeding is not recommended during treatment with Vericiguat.

✔️ Precautions & Warnings

Vericiguat should not be given to pregnant women due to the potential for fetal harm. A pregnancy test should be conducted in females of reproductive age before starting the treatment. Effective contraception should be used during treatment and for one month after discontinuation of the medication.

Store at a temperature below 30°C, away from light and moisture. Keep the medicine out of the reach of children.
