How does Klarix 250mg Tablet work?

Klarix 250 manages the growth of bacteria by interfering with its protein synthesis and binds specific proteins present in the bacterial ribosomes, thus stopping the growth of the bacteria inside the body, which helps to reduce the infection and its symptoms.

Side Effects of Klarix 250mg Tablet

Common side effects include:

• Nausea
• Vomiting
• Diarrhea
• Abdominal pain
• Stomatitis and glossitis
• Headaches
• Allergic reactions (ranging from mild skin reactions to anaphylaxis)
• Taste perversion
• Transient CNS effects (anxiety, dizziness, insomnia, hallucinations)

Quick Suggestions:

• Take it 1 hour before or two hours after food.
• Do not take antacids 2 hours before or after taking Claribid 500 Tablet.
• Try not to skip a dose. And if a dose is skipped, take it as soon as you remember, do not double the dose to compensate.
• Clarithromycin is used for treating bacterial infections and it is proven to be safe and effective. Clarithromycin is available in different dosage forms.

Indication of Klarix 250mg Tablet

Klarix is indicated for the treatment of the following conditions:

• Acute bacterial exacerbation of chronic bronchitis in adults
• Acute maxillary sinusitis
• Community-acquired pneumonia
• Pharyngitis or tonsillitis
• Uncomplicated skin and skin structure infections
• Acute otitis media in pediatric patients
• Treatment and prophylaxis of disseminated mycobacterial infections
• Helicobacter pylori infection and duodenal ulcer disease in adults

Pharmacology: Klarix 250mg Tablet

Clarithromycin works by inhibiting protein synthesis in susceptible bacteria. It binds to the donor site on the 50S subunit of the bacterial ribosome, blocking translocation. This antibiotic is effective against most Gram-positive bacteria, Chlamydia, some Gram-negative bacteria, and Mycoplasmas. Clarithromycin demonstrates equal or greater activity compared to Erythromycin against most Gram-positive bacteria and is more acid-stable, enhancing tolerance. It exhibits twice the activity of Erythromycin against Haemophilus influenzae, while most Gram-negative bacteria are resistant due to penetration issues.

Dosage & Administration: Klarix 250mg Tablet

Adults:

• Acute bacterial exacerbation of chronic bronchitis: 250-500 mg every 12 hours for 7-14 days.
• Acute maxillary sinusitis: 500 mg every 12 hours for 14 days.
• Community-acquired pneumonia: 250 mg every 12 hours for 7-14 days.
• Pharyngitis or tonsillitis: 250 mg every 12 hours for 10 days.
• Uncomplicated skin infections: 250 mg every 12 hours for 7-14 days.
• Mycobacterium avium disease: 500 mg every 12 hours until low risk for infection.

H. pylori Infection (Combination Therapy):

1. Triple Therapy: 500 mg clarithromycin, 30 mg lansoprazole, and 1 g amoxicillin every 12 hours for 10-14 days.
2. Triple Therapy: 500 mg clarithromycin, 20 mg omeprazole, and 1 g amoxicillin every 12 hours for 10-14 days, with an additional 14-18 days of omeprazole for ulcer healing if necessary.
3. Dual Therapy: 500 mg clarithromycin every 8 hours and 40 mg omeprazole once daily for 14 days, with an additional 14 days of omeprazole 20 mg for ulcer healing.

Children:

• 15 mg/kg/day divided every 12 hours for 10 days.
• For mycobacterial infections: 7.5 mg/kg every 12 hours, up to 500 mg.

Renal and Hepatic Impairment: 

Clarithromycin can be given without dosage adjustment in hepatic impairment with normal renal function. Adjust dosage or prolong intervals in severe renal impairment.

Klarix is effective in treating pharyngitis, pneumonia, sinusitis, otitis media, and skin infections in pediatric patients aged 6 months and older. Its safety for preventing disseminated Mycobacterium avium complex disease has been established in patients aged 20 months and older with advanced HIV. Its safety for children under 20 months has not been studied.

Interaction

Klarix is contraindicated when co-administered with:

• HMG-CoA reductase inhibitors (e.g., lovastatin, simvastatin)
• Gastroprokinetic agents (e.g., cisapride)
• Ergot alkaloids (e.g., ergotamine)
• Certain antipsychotics (e.g., pimozide)
• Antiarrhythmics (e.g., disopyramide, amiodarone)

Contraindications

Clarithromycin is contraindicated in patients with:

• Known hypersensitivity to clarithromycin, erythromycin, or other macrolide antibiotics.
• A history of cholestatic jaundice or hepatic dysfunction related to prior clarithromycin use.

Pregnancy & Lactation

Clarithromycin can be used in neonates and children at appropriate doses. Breastfeeding should be avoided while the mother is on this medication until treatment is completed. Its use during pregnancy is not recommended due to limited experience.

Precautions & Warnings

• Discontinue Klarix for severe hypersensitivity reactions.
• Avoid use in patients with known QT prolongation, ventricular arrhythmia, or significant bradycardia.
• Monitor for signs of hepatitis and consider the risk of increased mortality in patients with coronary artery disease.
• If diarrhea occurs, evaluate for Clostridium difficile-associated diarrhea (CDAD).
• Exacerbation of myasthenia gravis has been reported.

Storage Conditions:

Store in a cool, dry place, away from light and moisture. Use reconstituted suspension within 7 days at room temperature or 14 days when refrigerated. Keep out of reach of children.
