How does Lioresal 10mg Tablet work?

It works by inhibiting reflexes at the spinal level and acts as a CNS depressant with antinociceptive effects.

Side Effects of Lioresal 10mg Tablet

Common side effects include drowsiness, daytime sedation, dizziness, weakness, fatigue, headache, nausea, constipation, and urinary frequency. More severe effects like hallucinations, depression, respiratory depression, hypotension, and seizures can occur.

• Baclofen provides relief from rigidity, tension, and stiffness in muscles (spasticity) that may occur due to various conditions affecting the nervous system.
• Take it with or after food or a glass of milk.
• Baclofen may cause dizziness or sleepiness. Do not drive or do anything requiring concentration until you know how it affects you.
• Avoid consuming alcohol when taking Baclofen as it may cause excessive drowsiness.
• Inform your doctor if you are pregnant, planning to conceive, or breastfeeding.
• Do not stop taking it suddenly without talking to your doctor if you've been on it for a long time

Indications:

Lioresal 10mg Tablet is used to manage muscle spasms caused due to cerebral palsy, multiple sclerosis, stroke or due to any other nerve or spinal cord disorders.

Pharmacology

Baclofen inhibits both monosynaptic and polysynaptic reflexes at the spinal level by stimulating the GABAB receptors, which inhibit the release of glutamate and aspartate. It may also act at intraspinal sites producing CNS depression. Baclofen also exerts an antinociceptive effect.

Dosage & Administration of Lioresal 10mg Tablet

For adults and children over 10 years, the starting dose is 5 mg three times daily with food, gradually increasing to a maximum of 100 mg daily. 

For children under 10 years, the starting dose is 2.5 mg four times daily, with maintenance doses varying by age.

Interaction

Lioresal may interact with CNS depressants, alcohol, synthetic opiates, antihypertensives, tricyclic antidepressants, levodopa, carbidopa, MAO inhibitors, magnesium sulfate, and neuromuscular blocking agents. Always consult your doctor about potential interactions.

Contraindications

Lioresal is contraindicated in patients with hypersensitivity to any component of the product. It should be used cautiously in patients with renal impairment, psychiatric disorders, epilepsy, peptic ulcers, cerebrovascular diseases, and respiratory or hepatic failure.

Pregnancy & Lactation

Lioresal is a pregnancy category B3 drug. It should be used during pregnancy only if the potential benefits outweigh the risks. Baclofen is excreted in breast milk, but the quantities are generally too small to affect the infant.

Precautions & Warnings

• Lower doses (approximately 5 mg per day) should be used for patients with impaired renal function or those undergoing chronic hemodialysis.
• Patients suffering not only from spasticity but also from psychotic disorders, schizophrenia, depressive or manic disorders, or confusional states should be treated cautiously and closely monitored as exacerbations of these disorders may occur.
• In patients with epilepsy and muscle spasticity, Lioresal may be used under appropriate supervision provided that adequate anticonvulsive therapy is continued. Lowering of the convulsion threshold may occur and seizures have been reported after the cessation of Lioresal therapy or with overdose.
• Lioresal should be used with caution in patients with or with a history of peptic ulcers, cerebrovascular diseases, or hepatic, renal, or respiratory failure.
• Careful monitoring of respiratory and cardiovascular function is essential, especially in patients with cardiopulmonary disease and respiratory muscle weakness.
• During treatment with Lioresal, neurogenic disturbances affecting the emptying of the bladder may improve. However, in patients with preexisting sphincter hypertonia, acute retention of urine may occur. Lioresal should be used with caution in these circumstances.
• Lioresal has not significantly benefited patients with stroke. These patients have also shown poor tolerance to the medicine.
• Appropriate laboratory tests should be performed periodically in patients with hepatic diseases or diabetes mellitus to ensure that no medicine-induced changes in these underlying diseases have occurred.
