How does Finapark 100 work?

It works by increasing the amount of dopamine (a natural substance needed to control movement) in the brain. Thereby improving the symptoms of Parkinson’s disease.

Directions for Use:

Swallow Finapark 100 as a whole with water. Do not chew, crush, or break it. Your doctor will decide the correct dose and duration for you depending on your age, body weight, and disease condition.

Side Effects of Finapark 100

Common adverse reactions include:

• Dyskinesia
• Falls
• Nausea
• Insomnia

There is no human experience reported with Finapark overdose. If an overdose occurs, discontinue Finapark and administer supportive treatment as necessary, along with dietary restrictions of tyramine for several weeks.

Quick Suggestions:

• Safinamide can be taken with or without food or as directed by your doctor.
• Reduce/avoid a high-protein diet while you are taking the medicine as it may cause an unsafe rise in your blood pressure.
• Use caution while driving or doing anything that requires concentration as Safinamide can make you very sleepy.
• Talk to your doctor in case you develop hallucinations or other changes in your behavior.
• Do not stop taking Safinamide without first talking to your doctor. Sudden stopping may cause confusion, stiff muscles, and a high fever.

Indication of Finapark 100

Finapark is a monoamine oxidase type B (MAO-B) inhibitor indicated as an adjunctive treatment to levodopa/carbidopa for patients with Parkinson's disease who experience "off" episodes. It is not effective as monotherapy for Parkinson's disease.

Pharmacology

The exact mechanism of action of Safinamide Mesylate in Parkinson’s disease is not fully understood. It works by inhibiting monoamine oxidase B (MAO-B), which blocks the breakdown of dopamine, leading to increased dopamine levels and enhanced dopaminergic activity in the brain.

Dosage & Administration of Finapark 100

• Usual Initial Dosage: The recommended starting dose is 50 mg orally once daily, taken at the same time each day. If a dose is missed, take the next dose at the same time the next day.
• Maintenance Dosage: After 2 weeks, the dose may be increased to 100 mg once daily based on individual need and tolerability. If discontinuation is required, taper the dose to 50 mg for one week before stopping.
• Use in Children: Safety and effectiveness in infants and children have not been established.
• Geriatric Use: No significant differences in safety or effectiveness were observed between geriatric and younger patients.
• Hepatic Impairment: Plasma concentrations of Safinamide may be increased in patients with hepatic impairment.

Interaction

• Selective Serotonin Reuptake Inhibitors: Monitor for serotonin syndrome.
• Sympathomimetic Medications: Monitor for hypertension.
• Tyramine: Risk of severe hypertension.
• Substrates of Breast Cancer Resistance Protein (BCRP): Potential increase in plasma concentration of BCRP substrates.

Safinamide is contraindicated in patients with:

• Concomitant use of:
  • Other monoamine oxidase inhibitors or potent MAO inhibitors (e.g., linezolid)
  • Opioid drugs (e.g., tramadol, meperidine, and derivatives)
  • Selective norepinephrine reuptake inhibitors; tri- or tetra-cyclic antidepressants; cyclobenzaprine; methylphenidate, amphetamine, and their derivatives; St. John’s wort.
  • Dextromethorphan.
• A history of hypersensitivity to safinamide.
• Severe hepatic impairment (Child-Pugh C: 10-15).

Pregnancy & Lactation

• Pregnancy Category C: There are no adequate and well-controlled studies in pregnant women. Developmental toxicity, including teratogenic effects, was noted in animal studies. Hyperbilirubinemia-related skin discoloration was observed in rat pups indirectly exposed through lactation.
• Lactation: It's unknown if this drug is present in human milk. A decision should be made to either discontinue nursing or the drug, considering its importance to the mother.

Precautions & Warnings

• May cause or worsen hypertension.
• Risk of serotonin syndrome when used with MAO inhibitors, antidepressants, or opioids.
• Potential for falling asleep during daily activities.
• Possible exacerbation of dyskinesia; consider reducing levodopa dosage.
• Risk of hallucinations and psychotic behavior.
• This may lead to problems with impulse control/compulsive behaviors.
• Potential for withdrawal-emergent hyperpyrexia and confusion.
