Amenavir tablet is used for the treatment of herpes simplex infection, recurrent herpes simplex, and herpes zoster (shingles). Due to its novel mechanism of action, it is a useful alternative for patients who have developed resistance to conventional antiviral agents.
Herpes simplex: 200 mg orally once daily after meals.
Recurrent herpes simplex: A single 1200 mg dose after meals, taken within 6 hours of symptom onset.
Herpes zoster: 400 mg orally once daily after meals.
Pediatric use: Safety and efficacy in children have not been established.
Common: Headache, nausea, diarrhea, abdominal pain, skin reactions, increased urinary biomarkers
Serious: Thrombocytopenia, gingival bleeding, palpitations
Amenavir is metabolized mainly via CYP3A4:
Ketoconazole (CYP3A4 inhibitor): Increases Amenavir plasma levels.
Rifampicin (CYP3A4 inducer): Reduces Amenavir concentration and efficacy.
Midazolam: Plasma levels may be reduced when co-administered.
Warfarin: No clinically significant interaction observed.
